Džon Henkok (engl. John Hancock; Brejntri, 23. januar 1737 — Boston, Masačusets, 8. oktobar 1793) je bio trgovac, državnik i poznati patriota tokom Američke revolucije. Služio je kao predsednik Drugog Kontinentalnog kongresa i kao prvi i treći guverner Komonvelta Masačusets. Ostao je upamćen po velikom i kitnjastom potpisu na Deklaraciji nezavisnosti Sjedinjenih Država, čak toliko da je izraz „Džon Henkok“ u Sjedinjenim Državama postao sinonim za potpis.
Pre Američke revolucije, Henkok je bio jedan od najbogatijih ljudi u trinaest kolonija, nasledivši profitabilan trgovački biznis od svog ujaka. Henkok je započeo političku karijeru u Bostonu pod mentorstvom Samjuela Adamsa, uticajnog lokalnog političara, mada su se njih dvojica kasnije udaljili jedan od drugog. Kako su tenzije između kolonista i Velike Britanije počele da rastu tokom 1760-ih, Henkok je koristio svoje bogatstvo da podrži kolonijalne ciljeve. Postao je vrlo popularan u Masačusetsu, pogotovo nakon što su britanske vlasti zaplenile njegov slup Liberti 1768, i optužile ga za krijumčarenje. Ove optužbe su kasnije odbačene, ali se u istorijskim tekstovima Henkok ipak ponekad opisuje kao krijumčar, iako je tačnost tih navoda upitna.
Henkok je bio jedan od vođa iz Bostona za vreme krize koja je dovela do Američkog rata za nezavisnost 1775. Služio je više od dve godine kao delegat u Kontinentalnom kongresu u Filadelfiji, i kao predsedavajući Kongresa je bio prvi koji je potpisao Deklaraciju nezavisnosti. Henkok se vratio u Masačusets gde je izabran za guvernera Komonvelta, što je dužnost na kojoj je služio tokom većeg dela ostatka života. Iskoristio je svoj uticaj da osigura da Masačusets ratifikuje Ustav Sjedinjenih Država 1788.
